Tubulin Folding pathway outline:

Pathway I. Prefoldin mediated transfer of newly translated tubulin to Chaperonin Reaction 1. Association of prefoldin with newly translated tubulin Reaction 2. Transfer of tubulin from prefoldin to Chaperonin:ADP

Pathway II. Formation of tubulin folding intermediates by Chaperonin Reaction 1. Exchange of ADP for ATP in Chaperonin Reaction 2. Hydrolysis of ATP in the CCT:ATP:Tubulin complex leading to the Reaction 3. Release of tubulin folding intermediate (or unfolded tubulin ) which is Reaction 4. Recaptured by CCT:ADP (goto III)

Pathway III. Formation of the alpha-beta tubulin dimer Reaction 5. Capture of tubulin -alpha or beta folding intermediates by cofactors A-E (reversible rxn) Reaction 6. Association of (Alpha:E) and (beta:D) with CoC Reaction 7. Alpha:beta dimer formation: alpha:E+beta:D+CoC GTP->GDP --> release of the alpha:beta heterodimer

Pathway 1: Prefoldin mediated transfer of newly translated tubulin to Chaperonin Reaction 1: alpha/beta tubulin (defined set) + PFD → alpha/beta tubulin:PDF complex

Reaction 2: alpha/beta tubulin:PDF complex + CCT:ADP complex -> alpha/beta tubulin:PDF:CCT:ADP complex

Reaction 3.Dissociation of PFD from tubulin (no ATP/GTP hydrolysis) alpha/beta tubulin:PDF:CCT:ADP complex-> alpha/beta tubulin:CCT:ADP +PDF

Pathway 2: Formation of tubulin folding intermediates by Chaperonin Reaction 4.ADP is exchanged for ATP in the (ADP:CCT):(GTP:Tubulin) complex (ADP:CCT):(GTP:Tubulin) ->(ATP:CCT):(GTP:Tubulin)

Reaction 5. Conformational change in CCP and Tubulin GTP:Tubulin:CCP:ADP --> GTP:*Tubulin(folding intermediate):CCP*:ATP (how many molecules of ADP->ATP)

Reaction 6. GTP:Tubulin*(Quasi-native folding intermediate):CCP*:ATP-> GTP:*Tubulin(quasi-native folding intermediate) + CCP*:ADP +PPi

Pathway 3: Formation of the alpha-beta tubulin dimer Reaction 7. *Beta-tubulin:GTP + Cofactor A → *Beta-tubulin:GTP: Cofactor A(reversible)

Reaction 8. *Beta-tubulin(:GTP +Cofactor D → *Beta-tubulin:GTP: Cofactor D (reversible)

Reaction 9 *Beta-tubulin:GTP: Cofactor A+ Cofactor D → *Beta-tubulin:GTP: Cofactor D + Cofactor A (reversible)

.Reaction 10 *alpha-tubulin:GTP + Cofactor B -> *alpha-tubulin:GTP:Cofactor B(reversible)

Reaction 11. *alpha-tubulin:GTP + Cofactor E -> *alpha-tubulin:GTP:Cofactor E(reversible)

Reaction 12. *alpha-tubulin:GTP:Cofactor B + Cofactor E-> *alpha-tubulin:GTP:Cofactor E + Cofactor B (reversible)

Reaction 13. Beta-tubulin:GTP:Cofactor D+ alpha-tubulin:GTP:Cofactor E -> Beta-tubulin:GTP:Cofactor D:alpha-tubulin:GTP:Cofactor E

Reaction 14. Beta-tubulin:GTP:Cofactor D:alpha-tubulin:GTP:Cofactor E + Cofactor C-> Beta-tubulin:GTP:Cofactor D:alpha-tubulin:GTP:Cofactor E : Cofactor C

Reaction 15 .Beta-tubulin:GTP:Cofactor D:alpha-tubulin:GTP:Cofactor E : Cofactor C-> GDP:Beta-tubulin :alpha-tubulin:GTP heterodimer +Cofactor E+ Cofactor D+ Cofactor C+ PPi
